Abstract:
Lung cancer in man is today the leading cause of cancer death in the world. We have used transgenic mice with lung targeted expression of the CRaf kinase to evaluate genes frequently altered in human lung adenocarcinoma for their effect on tumor progression. A remarkable feature of the CRaf-induced lung tumors was the lack of tumor progression as judged from extensive histological examination. In a search for genes that modify tumor development we recently observed that bcl-2 is required for efficient lung tumor development in mice. Elucidation of additional genes that accelerate tumor development are likely to include, besides regulators of cell death, those genes that are involved in maintaining cell-cell contact and dissemination of cells to distant tissue regions.
