Several BMPs (bone morphogenetic proteins) have been reported to induce ectopic or orthotopic bone formation. Nevertheless, the BMPs from different subgroups activate and bind to receptor chains differently. They interact also differently with proteoglycans and inhibitors as chordin, noggin, a.o. Therefore, a detailed structure/function analysis (protein crystallography, X-ray analysis, mutation/interaction analysis) will be performed. In addition, the repertoire of osteogenic stem cells for receptor chains, signalling proteins and inhibitors will be elucidated (RT PCR, gene array). Thereby those interaction important for bone formation shall be identified and exploited for tissue engineering. Targets for therapeutic intervention in bone metabolism shall be found. The existing know how and equipment for protein analysis are offered for all collaborating groups.
