Molecular Mechanisms Controling NFATc1 Transcription in Lymphoid Cells in vivo

Abstract:

In TPC5 molecular mechanisms shall be determined which control the activity of the Nfatc1 locus in


peripheral T cells in vivo. We intend to investigate (1.) which remote regulatory DNA elements


control the Nfatc1 locus in vivo and (2.) which factors and mechanisms acting through (proximal)


promoter and (remote) enhancer/silencer elements regulate the induction, autoregulation and


repression of NFATc1/αA transcription in peripheral T cells, i.e. of a gene whose expression is under


control of a postive feedback mechanism. These experiments shall elucidate mechanisms controling


decisive steps of immunity or tolerance induction.
