Physiological BMP signals in mesenchymal stem cells: factors, chain of signalling receptors, modulators and signal proteins.

In model systems a series of BMPs (bone morphogenetic proteins) can induce the generation of ectopic as well as orthotopic bone. Recent data indicate, that BMP subtypes bind to different receptors, for which they display variable affinities. Besides that they variably interact with molecules of the matrix and the cell surfaces like for example proteoglycans, which modify BMP activity. Aim of the project is to characterize relevant protein interactions by 1) detailled analyses of structure and function (proteincristallography, x-ray structure analysis, Mutation-/Interaktion analyses), 2)by elucidating the expression profile of mesenchymal stem cells in terms of receptor chains, signalling proteins and modulating proteins. New data shall be applicated in Tissue engineering. An additional aspect is the development and the design of drugs which are capable of modulating bone modeling in therapeutic applications. The experience and tools established in the group for detailed biochemical analyses of proteins can be used by all projects.
