Abstract:
Materials (pseudomembrane, periprosthetic tissue) is stored at -80°C. After isolation of total RNA cDNA is generated by reverse transcription using super script II. By PCR analysis gene products representing osteoresorptive mediators are analyzed. Intensities of the PCR products obtained from about 20 samples are semiquantitatively analyzed by densitometry. Real time PCR is planned to quantitate differential expressions between RNA´s isolated from patients with septic or aseptic prosthetic loosening. Results of the project should allow for a better differential diagnosis of septic and aseptic loosening.
