Structural and functional characterization of the coronavirus 3C-like proteinase
Project management at the University of Würzburg:
Coronaviruses are mainly associated with respiratory and gastro-intestinal infections in both humans and animals. The expression and activation of the coronavirus replication complex involves a virus-encoded cysteine proteinase activity, which cleaves the replicase polyproteins at 11 conserved sites and also releases the key replicative proteins, such as the RNA-dependent RNA polymerase and helicase. The project aims at the comprehensive characterization of the 3C-like proteinase substrate specificity and the determination of its three-dimensional structure by X-ray crystallography (in collaboration with the IMB Jena). The information obtained in these studies will provide the basis for the development of proteinase assays suitable for high-throughput screening approaches to identify potential inhibitors from compound libraries and allow the rational design of selective proteinase inhibitors.
