Defective transcriptional activation in tumors of lymphatic tissues

Following activation by CD40, mature B lymphocytes express high levels of CD95 and are killed by T cells expressing CD95Ligand, unless their Antigen receptor (BCR) is also ligated. Protection by BCR ligation is partly mediated by induction of bcl-2 family members, but also by the induction of c-Flip which attenuates the death receptor induced signalling complex and the activation of the caspase cascade. c-Flip and A1, a bcl-2 family member, collaborate in rescuing B cells for survival. The project aims at studying transcription factors involved in c-FLIP activation, using luciferase reporter constructs. Functionally, the cooperative protective effects of A1 and c-Flip is studied in transgenic mice and in B cells transduced in vitro with A1 encoding viruses and mutants thereof.
