Project management at the University of Würzburg:
Differentiation of B-lymphocytes to immunoglobulin secreting plasma cells is regulated by the transcription factor Blimp-1. High levels of Blimp-1 up regulate Ig gene transcription and cell surface markers which are characteristic of plasma cells. On the other hand, c-myc and the antiapoptotic bcl-2 family member A1 are suppressed by Blimp-1, leading to a shortened life span of terminally differentiated plasma cells. With the help of A1-promoter-luciferase reporter constructs and mutants thereof we currently investigate, whether the A1 promoter is a direct or indirect target of Blimp-1 suppression.
