Specificity and activation of gd T cells of the rat: Importance for control of infectious diseases

In contrast to other species, rat T cells express the CD8ab heterodimer not only on MHC class I restricted ab T cells but also on gd T cells. The CD8ab heterodimers on both T cell populations showed no obvious differences in lck association and phosphorylation, suggesting that CD8ab on gd T cells could as an potential coreceptor in MHC class I restricted antigen recognition, but analysis of the gd TCR (CDR3d length and V gene usage) showed no differences between CD8 negative, CD8aa and CD8ab gd T cells. Interestingly, the CD8b chain on gd T cells but not on ab T cells was found to be modulated (protein and mRNA level) after TCR and CD28 engagment. This result may indicate a knew hitherto unknown function of CD8 on gd T cells.
Furthermore rat strain specific differences in the response of gd T cells against Toxoplasma gondii gd T cells were analysed and found to be inversely correlated with formation of brain cyst. The generation of a rat model of congenital Toxoplasmosis was started.
