The prognosis of malignant pediatric diseases has been improved substantially. About 70% of patients can be healed. Thus long term complications of anti-cancer therapy become more and more significant. One of the most important complications of cytostatic therapy in childhood is nephrotoxicity. Renal disease is significant for late cardiovascular morbidity and mortality. It also is important because many cytostatic drugs are eliminated by the kidneys.
The aim of the study is to determine long-term complications of anti-cancer therapy. The tubulo-toxicity of Ifosfamide will be a major part of the project. Ifosfamide causes renal wasting of phosphate, sodium, HCO3, aminoacids and sometimes water. The most severe disease is secondary Fanconi- syndrome. The project´s goals are to study the severity of phosphat wasting under Ifosfamide and the significance of phosphate wasting for bone metabolism and growth. Furthermore cellular and molecular mechanisms of tubulo-toxicity and phosphate wasting by the kidneys will be studied in an established in vitro-model.
