Architecture of the nuclear periphery of meiotic cells

In a previous project, we were able to show that in Lmna-/- mice lacking A-type lamins, spermatogenesis is disrupted and that spermatocytes show severe defects in synaptic pairing of chromosomes. These results provided evidence for the involvement of the nuclear lamina in meiotic nuclear events and that A-type lamins are an important determinant of male fertility. Within the frame of the present project we propose to continue with the characterization of the molecular architecture of the nuclear periphery of mammalian meiotic cells. Lamin C2-interacting proteins should be identified and characterized. The role of the second germ line-specific lamin (B3) should be investigated using knock out mice. As proposed for lamin C2, lamin B3 interacting proteins should be identified and characterized.
