Sonderforschungsbereich 355: Pathophysiologie der Herzinsuffizienz



Zentrales Thema des SFB 355 sind die Ursachen, Folgen und Behandlung der Herzinsuffizienz. Die chronische Herzinsuffizienz kann das Endstadium verschiedener kardiovaskulärer Grunderkrankungen wie koronare Herzkrankheit, Herzinfarkt, Hypertonie u.a. sein und hat aufgrund ihrer zunehmenden Häufigkeit eine große medizinische und sozioökonomische Bedeutung. Die multifaktorielle Genese, vor allem aber die Konsequenzen der Herzinsuffizienz für alle Organe erfordert einen multidisziplinären Forschungsansatz. Im Projektbereich A (Kernspinresonanz) des SFB 355 werden neue, auf der Kernspinresonanztechnik basierende Methoden zur Analyse von Herzanatomie, -funktion, -durchblutung und -stoffwechsel entwickelt und sowohl experimentell zur Erforschung der pathophysiologischen Vorgänge bei Herzinsuffizienz als auch klinisch an Patienten mit Herzversagen angewendet. Im Projektbereich B (Pathophysiologie) werden grundlegende Untersuchungen zu den Mechanismen der nach Myokardinfarkt chronisch auftretenden Herzinsuffizienz durchgeführt, insbesondere wird die Rolle körpereigener Hormone (Endothelin, Atriale Faktoren, Stickstoffmonoxid etc.), wichtiger Komponenten der Calcium-Homoöstase und der Endothelzellen untersucht. Der Projektbereich C (Pathobiochemie und Molekularbiologie) ist zell- und molekularbiologisch ausgerichtet, wobei die Analyse der Funktion von Hormon-, Adhäsions- und Wachstumsfaktor-Rezeptoren und ihre Bedeutung für die Herzinsuffizienz im Vordergrund stehen. Von großer Bedeutung sind hier genetisch bedingte Tiermodelle (transgene Mäuse) von Herzerkrankungen, die mit den modernsten Methoden der Biochemie, Pharmakologie, Physiologie und Physik untersucht werden.
